Abstract
This study was focused on the aeration effect on thermostable T1 lipase production and fermentation performance by recombinant E. coli. Observation of fermenter aeration strategies such as impeller speed, airflow rate, and dissolved oxygen tension (DOT) throughout the fermentation was carried out and compared. 89.82 U/ml of lipase activity was observed at 1.5 L/min (1 vvm) airflow rate with 250 rpm impeller speed. When impeller speed increased from 250 to 350 rpm, an increase in lipase activity up to 93.03 U/mL was observed. No improvement in lipase production was observed at controlled DOT. More studies on aeration effect needed in order to improve higher lipase production.
